 Mutant lines Method
  Plant Materials
 
  • Lotus japonicus MG-20 (Miyakojima)

  EMS treatment
 
  • Rub seed's surface with sandpaper → absorption of water → add seeds in 0.4% (v/v) EMS solution → over night → wash seeds for 6 to 8 hrs → saw seeds on the peat moss.

  Growth Condition
 
  • 23℃ 16h(light):8h(dark)
  • After screening, seedlings are planted on soil (5cm in diameter).

  M1 lines
 
  • About 3000 lines

  Medium
 
  • 1/2 Okada and Shimura medium with 1.5% agar (OS medium : Aust J Plant Physiol 1992: 19, 439-448).
  • Peat moss (Sakata seeds co., Japan)

  Screening on the agar medium
 
  • Observing the form of one- to two-weeks-old seedlings on the 1/2 OS medium with 1.5% agar.
  • About 20 seedlings were observed for each M1 line.

  Screening on the peat moss
 
  • Observing the form of two- to three-weeks-old seedling.
  • About 30 seedlings were observed for each M1 line.

  Observing the form of the adult phenotype
 
  • After observations on the agar medium and the peat moss, one to three seeding were transplanted on soil.
  • Observing the phenotype of two to three months old plants.
